Quantificare l'attivazione dell'inflammasoma senza noiose fasi di lavaggio

Il Lumit™ IL-1β Immunoassay misura quantitativamente l'IL-1β rilasciata in campioni di colture cellulari utilizzando un protocollo semplice e senza lavaggio. È sufficiente aggiungere gli anticorpi marcati al campione, aggiungere il reagente di rilevazione e leggere il segnale luminescente utilizzando un luminometro standard a lettura di piastra. L'intero protocollo viene completato in meno di 70 minuti! Il test può essere utilizzato direttamente sulle cellule in coltura o sul terreno di coltura trasferito in una piastra di analisi separata.

Come funziona il Lumit™ IL-1β Immunoassay

Gli anticorpi primari contro l'IL-1β sono stati selezionati per il loro rilevamento specifico e sensibile e sono stati marcati con le subunità LgBiT e SmBiT della NanoBiT® Luciferase. In presenza di IL-1β, le subunità si uniscono per formare un enzima luciferasi attivo. L'aggiunta di un substrato ottimizzato genera un segnale luminescente proporzionale ai livelli di IL-1β.

Il formato add-and-read del Lumit™ IL-1β Immunoassay consente uno screening ad alto rendimento per i modulatori del rilascio di IL-1β.

IL-1b inhibitor high throughput screening — L'inibitore dell'inflammasoma NLRP3, MCC950, impedisce l'attivazione della caspasi-1 e il rilascio di IL-1 maturaβ. Una titolazione di MCC950 è stata aggiunta a cellule THP-1 differenziate (5 ×^104/pozzetto) seguite da LPS (20 EU/ml) o da un veicolo. L'immunodosaggio Lumit™ IL-1β (Human) è stato eseguito dopo 5 ore.
