What is NanoPrism™ luciferase technology?

Complex biological systems require monitoring multiple processes to understand true cause-and-effect relationships and validate experimental results. Traditional single-readout assays only tell part of the story. NanoPrism™ technology adds a second bioluminescent readout to your existing NanoLuc®/NanoBiT® assays, giving you all the benefits of live-cell bioluminescence for both signals in a single sample.

Key capabilities:

  • Dual-signal detection with a single substrate addition
  • Seamless integration with existing NanoLuc® or NanoBiT® workflows
  • Live-cell compatible for real-time monitoring
  • Sensitive to endogenous expression levels

How does NanoPrism™ luciferase technology work?

NanoPrism™ technology was engineered by incorporating circularly permuted NanoLuc (PrismaLuc) or LgBiT (PmLgBiT) into HaloTag. PrismaLuc™/PrismaBiT™ luciferases generate a spectrally distinct bioluminescent signal through energy transfer to a covalently bound fluorophore (e.g., Janelia Fluor® 549 HaloTag® ligand; JF549), enabling quantitative measurement of two biological events when combined with NanoLuc®/NanoBiT® luciferases in the same sample. This dual-color capability works on standard plate readers and bioluminescence imagers using filtered luminescence detection.

Applications of NanoPrism™ technology

Ideal for researchers studying:

  • Targeted protein degradation
  • Biased signaling
  • Protein-protein interactions
  • Multi-cell type dynamics
  • And more!
