La generazione corretta di un file di calibrazione spettrale è fondamentale per valutare i sistemi multicolore. Lo standard di matrice GenePrint® 5C contiene frammenti di matrice marcati con cinque coloranti fluorescenti: Fluoresceina, JOE, TMR-ET, CXR-ET e WEN. Una volta generato, il file di calibrazione spettrale viene applicato durante la raccolta dei dati PowerPlex® per calcolare e compensare la sovrapposizione spettrale tra i diversi colori dei coloranti fluorescenti.
