Fast Quantification of Common Neosubstrates for CRBN-Based Degraders

Lumit® Immunoassay Cellular Systems for Neosubstrates enable rapid measurement of 5 common CRBN neosubstrates (GSPT1, IKZF1, IKZF2, IKZF3, and SALL4) directly in cell lysates. No cell line engineering is required. Optimized antibody formulations simplify adaptation across cell lines of interest. Researchers can quickly assess both on-target degradation and off-target neosubstrate liabilities in the cellular contexts most relevant to their therapeutic application.

Why is neosubstrate selectivity critical in CRBN-based degrader development?

Targeted protein degradation (TPD) harnesses cellular machinery to eliminate disease-relevant proteins. CRBN-based degraders have achieved the greatest clinical success to date. A key challenge is characterizing neosubstrate degradation specificity, the proteins that become degradation targets through degrader-induced conformational changes to CRBN. For PROTACs, neosubstrates are off-target liabilities. For molecular glues, they are the intended targets. Either way, selectivity matters, since non-selective degradation of multiple neosubstrates poses safety risks. Protein expression and cellular context vary by cell type and disease state. This makes profiling in disease-relevant cell lines essential.

How the Assay Works

The Lumit® Immunoassay Cellular System does not require lysate transfer or wash steps, providing a simple, single-plate workflow. After treatment with degraders, cells are lysed, and an antibody mixture is added to detect the target analyte. The antibody mixture consists of two primary antibodies specific to the target and the matching SmBiT- and LgBiT-conjugated secondary antibodies. In the presence of the analyte, the antibodies come into close proximity, allowing SmBiT and LgBiT to form an active luciferase and generate a bright luminescence signal upon addition of detection reagent. Degradation of the target analyte will lead to a loss of signal.

Quantify neosubstrate levels across diverse cell lines with Lumit® Immunoassays. Molecular glue-mediated degradation of GSPT1 (Panel A) and IKZF1 (Panel B) was assessed in various non-engineered cell lines after 3 hours or 24 hours of compound treatment. Degradation potencies and Dmax differed significantly across all cell lines.

Monitor Off-Target Degradation of Common Neosubstrates by CRBN Degraders

Evaluate specificity of CRBN degraders with Lumit® Immunoassays for CRBN Neosubstrates. The Lumit® Immunoassays for 5 common CRBN Neosubstrates are used to measure neosubstrate degradation by CRBN-based PROTACs targeting BTK/IKZF1/IKZF3 (left) or STAT3 (right). On-target degradation of IKZF1 and IKZF3, and partial off-target degradation of SALL4, were observed after treatment with the dual-modality BTK/IKZF1/IKZF3 PROTAC dd-03-171 for 3 hours. No off-target degradation was observed for the STAT3 PROTAC SD-36 following 24 hours of treatment. The following cell lines were used for profiling: HEK293 (GSPT1), Jurkat (IKZF1 and IKZF2), MM.1S (IKZF3), PA-1 (SALL4), Ramos (BTK) and K562 (STAT3).
