Achieve Successful Transfections with Less Effort
Quality-controlled manufacturing procedures ensure TransfectNow™ HEK293 Cells undergo consistent and high-efficiency transfections that are comparable to optimally cultured and harvested HEK293 cells.
TransfectNow™ HEK293 Cells are comparable to cultured HEK293 cells. Six Kinase-NanoLuc® fusion vector constructs were individually transfected in TransfectNow™ HEK293 Cells and freshly harvested HEK293 cells using FuGENE® HD Reagent. Each set of transfected cells was assayed using the NanoBRET® TE Intracellular Kinase Assay K-10 with or without a competing saturating dose of inhibitory compound (CC1 pan-kinase inhibitor) to compete with the BRET signal to determine the assay window. Total luminescence and BRET assay window were determined.
TransfectNow™ HEK293 Cells show day-to-day transfection efficiency. On two separate days, six Kinase-NanoLuc® fusion vector constructs were individually transfected in TransfectNow™ HEK293 Cells using FuGENE® HD Transfection Reagent. Each set of transfected cells was assayed using the NanoBRET® TE Intracellular Kinase Assay K-10 with or without a competing saturating dose of inhibitory compound (CC1 pan-kinase inhibitor) to compete with the BRET signal to determine the assay window. Total luminescence and BRET assay window were determined.
Successfully Transfect a Range of Fusion Vector Constructs
Comparable expression levels and BRET-based assay windows were obtained for 192 individual Kinase-NanoLuc® Fusion Vectors transfected using TransfectNow™ HEK293 Cells or freshly harvested HEK293 cells.
Cells transfected with Kinase-NanoLuc® fusion vectors were assayed using the NanoBRET® TE Intracellular Kinase Assay K-10 with or without a competing saturating dose of inhibitory compound (CC1 pan-kinase inhibitor) to determine BRET assay window. Panel A. Correlation plot shows that comparable NanoLuc® expression levels were obtained for all transfections with TransfectNow™ HEK293 Cells and freshly harvested HEK293 cells. Panel B.
