Quantificare il rilascio di citochine senza le noiose fasi di lavaggio
Il Lumit™ IL-6 Immunoassay misura quantitativamente l'IL-6 rilasciata in campioni di colture cellulari utilizzando un protocollo semplice e senza lavaggio. È sufficiente aggiungere gli anticorpi marcati al campione, aggiungere il reagente di rilevazione e leggere il segnale luminescente utilizzando un luminometro standard a lettura di piastra. L'intero protocollo viene completato in meno di 70 minuti! Il test può essere utilizzato direttamente sulle cellule in coltura o sul terreno di coltura trasferito in una piastra di analisi separata.
Come funziona l'immunodosaggio Lumit™ IL-6
Gli anticorpi primari contro l'IL-6 sono stati selezionati per il loro rilevamento specifico e sensibile e sono stati marcati con le subunità LgBiT e SmBiT della NanoBiT® Luciferase. In presenza di IL-6, le subunità si uniscono per formare un enzima luciferasi attivo. L'aggiunta di un substrato ottimizzato genera un segnale luminoso luminescente proporzionale ai livelli di IL-6.

Rilevazione rapida del rilascio di IL-6 dalle PBMC
Il Lumit™ IL-6 (Human) Immunoassay (a sinistra) e l'ELISA standard (a destra) sono stati utilizzati per quantificare il rilascio di IL-6 da PBMC trattate con tre induttori (cocktail di stimolazione cellulare, lipopolisaccaride o R848) per un massimo di 72 ore. I risultati sono simili, mentre l'immunodosaggio Lumit™ è stato completato in un quinto del tempo rispetto all'ELISA, senza necessità di trasferire e lavare le piastre o di lavarle.
