Il sistema Autophagy LC3 HiBiT Reporter Assay utilizza un reporter LC3 luminescente per misurare quantitativamente il flusso autofagico. Le cellule HEK293 Autophagy LC3 HiBiT Reporter sono state piastrate in piastre da 96 pozzetti completamente bianche a 20.000 cellule/pozzetto. Dopo che le cellule si sono fissate per una notte, è stato eseguito un trattamento in triplicato per i tempi indicati con concentrazioni crescenti di un induttore di autofagia di riferimento, PP242. Nano-Glo® HiBiT Lytic Reagent è stato aggiunto e la luminescenza è stata misurata dopo 10 minuti. Le cellule U2OS Autophagy LC3 HiBiT Reporter sono state piastrate in piastre da 96 pozzetti completamente bianche a 8.000 cellule/pozzetto.  Dopo aver fissato le cellule per una notte, le cellule sono state trattate con concentrazioni crescenti di un inibitore di riferimento, Baf A1, senza o con 2μM PP242 per 21 ore. Il segnale luminescente è stato normalizzato rispetto al controllo non trattato per ogni studio. Il trattamento con PP242 ha aumentato significativamente l'autofagia e diminuito il segnale luminescente per consentire una maggiore capacità di aumento del segnale mediante il cotrattamento con l'inibitore dell'autofagia, Baf A1.
